Shawn R. Campagna
Professor, Scientific Director UT-ORII, Faculty Director BSMMSC
Dr. Shawn Campagna completed his B.S. in Chemistry at North Carolina State University in 2000 and conducted research with Prof. Jonathan S. Lindsey on the chemical synthesis of bacterial chromosomes. He received his Ph.D. from Princeton University in 2006, after working with Prof. Martin F. Semmelhack on a joint project with Profs. Bonnie L. Bassler and Frederick M. Hughson to characterize the chemical properties of an inter-species bacterial signaling molecule, autoinducer-2. Campagna then performed post-doctoral fellow research with Prof. Joshua D. Rabinowitz at the Lewis-Sigler Institute for Integrative Genomics at Princeton University where he developed mass spectrometric methods for the identification of novel biochemical pathways and natural products from whole cell extracts. He joined the Chemistry Department at UT Knoxville in August 2007.

Research
My research program utilizes bioanalytical mass spectrometry and organic synthesis to both develop analytical tools and to probe biological phenomena, with a specific focus on understanding signaling systems and their links to metabolism. My lab has established collaborations with a number of other researchers, both at UTK and at other universities. By utilizing interdisciplinary environments both within my lab and among collaborators, the students in my lab are able to experience differing scientific perspectives. The organic group in the Campagna lab focuses on the synthesis of biologically relevant compounds, The largest of these initiatives is an ongoing collaboration with Dr. Jason Collier at Pennington Biomedical Research Center, aiming to design and synthesize novel glucocorticoid analogues that maintain their anti-inflammatory properties while minimizing their potential to induce unwanted side effects, such as steroid induced diabetes. This project is further enhanced with computational experiments, including Molecular Dynamic simulations and de novo drug design to better understand the structure-activity relationship between these ligands and their target protein, the glucocorticoid receptor. We have also started an exciting and highly collaborative medicinal chemistry project to develop novel radioconjugates for PET imaging and targeted cancer therapy. While the analytical group is involved in a wide array of collaborations, one of the main focuses has been to understand the small molecule produced by the gut microbiome as these regulate our overall health. The small molecule profile produced by the microorganisms that inhabit our bodies is highly individualized and sensitive to intrinsic (age, sex, ethnicity) and extrinsic factors (diet, geography, lifestyle), making it an ideal target in precision medicine. In addition, our lab is investigating the mechanisms behind quorum sensing, a cell-to-cell communication process that bacteria use to coordinate their behaviors. These multidisciplinary projects require the synthesis of novel metabolites and the analysis of mixture of molecules to understand how bacterial metabolism is regulated by these fascinating organisms.
